Biological Background: PI3 Kinase p85 alpha Function and Localization
- Encoded by the PIK3R1 gene, phosphoinositide 3-kinase regulatory subunit alpha (p85α) is the predominant regulatory isoform of class IA PI3 kinases, existing as multiple splice variants including isoform 1 (canonical), isoform 2 (brain/skeletal muscle), and isoform 4.
- Functions as an adaptor protein, using its SH2 domains to bind phosphorylated tyrosine residues on activated receptor tyrosine kinases (e.g., FGFR1-4, KIT, PDGFRA/B) and insulin receptor substrates, thereby recruiting the p110 catalytic subunit to the plasma membrane for local PIP3 production. Related references: PMID:7518429 PMID:17626883 PMID:19805105
- Essential for insulin-mediated glucose uptake and glycogen synthesis; it also modulates the ER stress response by promoting nuclear translocation of XBP1 isoform 2 in hepatocytes, contributing to improved glucose tolerance. Related references: PMID:20348923
- Mediates integrin beta-2 (ITGB2) signaling, coupling adhesion events to PI3K activation and cytoskeletal rearrangement.
- The protein is predominantly cytosolic and translocates to membrane microdomains upon stimulation; it contains an N-terminal SH3 domain, a proline-rich breakpoint cluster region homology domain, and two C-terminal SH2 domains.
- Post-translational modifications include phosphorylation on multiple tyrosine/serine residues (critical for activity and protein interactions), acetylation, and ubiquitination, which fine-tune its stability and signaling output.
- Clinically, germline or somatic variants in PIK3R1 are associated with SHORT syndrome (short stature, hyperextensibility, ocular depression, Rieger anomaly, teething delay) and various cancers, highlighting its role in growth and metabolic homeostasis.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a region (aa 240-380) within the inter-SH2 domain, which is conserved across human, mouse, and rat, consistent with the reported cross-reactivity.
- For Western blotting, a band at approximately 84 kDa is expected; consider using whole-cell lysates from insulin-stimulated HepG2 or 3T3-L1 cells to enhance detection.
- In IHC-P, antigen retrieval with citrate buffer (pH 6.0) is suggested; test optimal antibody dilution on formalin-fixed paraffin-embedded sections of human breast or colon carcinoma.
- ELISA suitability indicates reactivity with native or recombinant p85α; validate in your specific assay format with appropriate positive and negative controls.
