Biological Background: PI3 Kinase p110 delta Function and Localization
- PI3K p110 delta is the catalytic subunit of class IA PI3K, encoded by the PIK3CD gene. It phosphorylates phosphatidylinositol 4,5-bisphosphate (PIP2) to generate phosphatidylinositol 3,4,5-trisphosphate (PIP3), a critical second messenger in signal transduction. Related references: PMID:15135396
- It plays essential roles in immune cell signaling, including B-cell receptor (BCR) and T-cell receptor (TCR) pathways, regulating lymphocyte proliferation, differentiation, and migration.
- Involved in both innate and adaptive immunity, p110 delta contributes to neutrophil chemotaxis, natural killer (NK) cell development and cytotoxicity, and mast cell degranulation.
- The protein is predominantly expressed in leukocytes, with the highest levels in peripheral blood mononuclear cells, spleen, and thymus, and low levels in testes, uterus, colon, and small intestine (PubMed:9235916). Isoform 2 is overexpressed in some colorectal and ovarian tumors. Related references: PMID:9235916
- Subcellularly, PI3K p110 delta localizes to the cytoplasm, and its kinase activity is regulated by post-translational modifications, including phosphorylation.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fragment spanning amino acids 170–300 of human p110 delta. Researchers should verify reactivity in their specific sample system, particularly when working with isoform 2, which may share epitopes.
- For western blotting, a band around 119 kDa is expected; consider using leukocyte or lymphoid tissue lysates as positive controls.
- For ELISA applications, optimization of coating and detection conditions is recommended. Cross-reactivity with mouse and rat orthologs may facilitate translational studies, but validation in the relevant species is advised.
