Biological Background: β-Catenin (CTNNB1) Function and Localization

  • Acts as a key downstream component of the canonical Wnt signaling pathway. In the absence of Wnt, forms a destruction complex with AXIN1, APC, and GSK3B that phosphorylates N-terminal Ser/Thr residues, leading to ubiquitination and proteasomal degradation. Upon Wnt stimulation, accumulates in the nucleus and serves as a coactivator for TCF/LEF transcription factors to activate Wnt-responsive genes.
