- NTRK1 encodes the high affinity nerve growth factor receptor, also known as TrkA, gp140trk, and p140-TrkA.
- TrkA is a receptor tyrosine kinase that binds neurotrophins, playing a critical role in neuronal survival, differentiation, and growth.
- The antibody specifically recognizes phosphorylation at tyrosine residues Y674 and Y675 of TrkA (isoform TrkA-I, P04629-2) and the analogous phosphorylation sites Y706 and Y707 of TrkB (Q16620).
- TrkA exists as multiple isoforms; isoform TrkA-I (used as immunogen reference) has a molecular weight of approximately 87 kDa, while the observed protein weight of the antibody includes additional bands due to glycosylation and potential cleavage products (35 kDa, 53-60 kDa, 77 kDa, 83-93 kDa).
- TrkB (NTRK2) is a closely related neurotrophin receptor that shares structural and phosphorylation‐site homology with TrkA.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic phosphorylated peptide corresponding to residues around Y674/Y675 of human TrkA. Verify reactivity and specificity in the relevant sample system, such as NGF‐stimulated neuronal cell lines.
- For WB, consider using phospho‐specific and total protein controls to confirm band identity; a band at ~87 kDa (full‐length TrkA) and lower molecular weight bands may be observed.
- For ELISA, optimize coating conditions with phosphorylated versus non‐phosphorylated peptides to assess phospho‐selectivity.
- Since the antibody also recognizes TrkB phosphorylation, validate cross‐reactivity by knockdown or using specific ligands (e.g., BDNF for TrkB activation).
