Biological Background: TrkA/NTRK1 Function and Localization
- Member of the neurotrophic tyrosine kinase receptor family; high affinity receptor for nerve growth factor (NGF) and also binds neurotrophin-3 (NTF3).
- Upon NGF binding, undergoes dimerization and autophosphorylation at tyrosine residues including Y490, creating docking sites for adaptor proteins such as SHC1 and FRS2.
Related references: PMID:1281417 - Activates multiple downstream signaling pathways including Ras-MAPK (via SHC1-GRB2-SOS) and PI3K-AKT1 (via SHC1 and SH2B1), promoting neuronal differentiation and survival.
Related references: PMID:15488758, PMID:17196528 - Isoform TrkA-III lacks NGF responsiveness, constitutively activates AKT1 and NF-kappa-B, and exhibits oncogenic activity when overexpressed in certain contexts.
- Localizes to the cell membrane and intracellular endosomes (early, late, recycling), cycling in response to ligand stimulation.
- Expressed primarily in neuronal cells (TrkA-II) and neural progenitors (TrkA-III), with TrkA-I found in non-neuronal tissues.
- Post-translational modifications include phosphorylation (key for kinase activity and signal transduction) and glycosylation; classified as a glycoprotein and phosphoprotein.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic phosphopeptide corresponding to phosphorylated Y490 of human TrkA; validate antibody specificity using non-phosphorylated controls or phosphatase treatment.
- For Western blot, an expected band near 87 kDa is observed in lysates from NGF-stimulated SH-SY5Y or PC12 cells; consider using a loading control.
- The antibody is applicable in ELISA; titrate conditions for optimal signal-to-noise ratio in your assay format.
- Cross-reactivity with mouse and rat is reported; verify performance in your target species' tissue or cell lines.
- Shipping on ice bag; store at -20°C upon receipt and avoid repeated freeze-thaw cycles.
