Biological Background: TrkA/NTRK1 Function and Localization

  • Member of the neurotrophic tyrosine kinase receptor family; high affinity receptor for nerve growth factor (NGF) and also binds neurotrophin-3 (NTF3).
  • Upon NGF binding, undergoes dimerization and autophosphorylation at tyrosine residues including Y490, creating docking sites for adaptor proteins such as SHC1 and FRS2.

  • Activates multiple downstream signaling pathways including Ras-MAPK (via SHC1-GRB2-SOS) and PI3K-AKT1 (via SHC1 and SH2B1), promoting neuronal differentiation and survival.
