Biological Background: TBK1 Function and Localization
- TBK1 is a serine/threonine-protein kinase encoded by the TBK1 gene, also known as NAK or T2K.
- It plays an essential role in regulating inflammatory responses to foreign agents by phosphorylating interferon regulatory factors (IRFs) IRF3 and IRF7, leading to their dimerization and nuclear translocation to induce antiviral genes such as IFNA and IFNB (PubMed:10581243, PubMed:11839743, PubMed:12692549, PubMed:12702806, PubMed:14703513, PubMed:15367631, PubMed:25636800). Related references: PMID:10581243, PMID:11839743, PMID:12692549, PMID:12702806, PMID:14703513, PMID:15367631, PMID:25636800
- TBK1 can also function as an NF-kappa-B effector by phosphorylating NFKBIA, IKBKB, or RELA, thus promoting NF-kappa-B nuclear translocation (PubMed:10783893, PubMed:15489227). Related references: PMID:10783893, PMID:15489227
- It acts in antibacterial autophagy by phosphorylating OPTN on Ser-177, enhancing LC3 binding (PubMed:21617041). Related references: PMID:21617041
- TBK1 positively regulates mTORC1 by phosphorylating MTOR, but also limits mTORC1 via RPTOR phosphorylation (PubMed:29150432, PubMed:31530866). Related references: PMID:29150432, PMID:31530866
- Subcellular localization is predominantly cytoplasmic.
- Ubiquitously expressed with higher expression in testis; also found in ganglion cells, nerve fiber layer, and microvasculature of the retina.
- Associated with diseases including amyotrophic lateral sclerosis and glaucoma; phosphorylation plays a critical role in its kinase activity and signaling.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic phosphorylated peptide surrounding Ser172 of human TBK1. For phospho-specificity validation, consider phosphatase treatment of samples or pre-incubation with non-phosphorylated competitor peptide.
- For western blot, the predicted molecular weight is approximately 84 kDa; ensure adequate separation and transfer, and use a molecular weight marker.
- IHC-P may require optimized antigen retrieval protocols; test both citrate and EDTA-based methods, and consider using phosphorylated antigen-friendly conditions.
- This rabbit polyclonal antibody is validated for ELISA; perform appropriate titration to determine optimal working concentration.
- Cross-reactivity with mouse and rat has been reported, but sequence conservation suggests it may also recognize other species; validate in your specific model system.
- As with all polyclonal antibodies, lot-to-lot variability can occur; consider validating each new lot in a pilot experiment.
