Biological Background: TBK1 Function and Localization
- Serine/threonine-protein kinase TBK1 plays an essential role in regulating inflammatory responses to foreign agents by phosphorylating IRF3 and IRF7, leading to transcriptional activation of pro-inflammatory and antiviral genes including type I interferons. Related references: PMID:10581243, PMID:11839743, PMID:12692549, PMID:12702806, PMID:14703513, PMID:15367631, PMID:25636800
- TBK1 forms dynamic signaling complexes with adapter proteins such as MAVS, STING1, TICAM1, TANK, and TBKBP1/SINTBAD, enabling context-dependent activation of IRF3 and NF-κB pathways. Related references: PMID:23453971, PMID:23453972, PMID:23746807, PMID:21931631, PMID:25636800, PMID:30842653, PMID:37926288
- Beyond its role in innate immunity, TBK1 regulates autophagy by phosphorylating OPTN on Ser177 to enhance antibacterial autophagy, and by phosphorylating SMCR8 and ATG8 family proteins to promote autophagosome maturation. Related references: PMID:21617041, PMID:27103069, PMID:31709703
- TBK1 can also activate NF-κB signaling by phosphorylating NFKBIA, IKBKB, or RELA, thereby contributing to nuclear translocation of NF-κB and supporting a wide range of immune and inflammatory responses. Related references: PMID:10783893, PMID:15489227
- The kinase participates in antiviral defense by attenuating retroviral budding via VPS37C phosphorylation and by controlling herpes virus HSV-1 and HSV-2 infections in the central nervous system. Related references: PMID:21270402, PMID:22851595
- TBK1 is ubiquitously expressed with higher levels in testis and is localized to the cytoplasm, consistent with its roles in signaling cascades and post-translational modifications.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic phosphorylated peptide around Ser172 of human TBK1, ensuring phosphorylation-specific detection. Include phosphatase inhibitors during lysis and sample preparation for WB.
- Confirm antibody specificity by comparing signals from phosphorylated and non-phosphorylated controls (e.g., lambda phosphatase treatment).
- The antibody cross-reacts with mouse TBK1; validate performance in your target species and tissue.
- For ELISA, optimize coating antigen and antibody dilution using phosphorylated peptide or cell lysates from stimulated samples.
- Consider using this antibody in combination with total TBK1 antibodies to assess phosphorylation changes.
