Biological Background: Tau (Phospho-T212) Function and Localization

  • Microtubule-associated protein tau (MAPT) is a neuronal phosphoprotein that promotes microtubule assembly and stability. Multiple isoforms are generated by alternative splicing, with short isoforms allowing cytoskeletal plasticity and longer isoforms stabilizing microtubules.

  • Located in the cytoplasm, cytosol, cell membrane, cytoskeleton, axons, and dendrites, and can be secreted. Predominantly expressed in neurons, with isoform PNS-tau in the peripheral nervous system and others in the central nervous system.
