Biological Background: Microtubule-associated protein tau (Tau, MAPT) Function and Localization

  • Microtubule-associated protein tau (MAPT) promotes microtubule assembly and stability, and is critical for establishing and maintaining neuronal polarity (PubMed:21985311). It acts as a linker between axonal microtubules and neural plasma membrane components (PubMed:21985311, PubMed:32961270). Related references: PMID:21985311 PMID:32961270
  • Tau exists in multiple isoforms generated by alternative splicing; short isoforms confer cytoskeletal plasticity, while longer isoforms stabilize microtubules.
  • Subcellularly, tau is predominantly cytoplasmic and cytoskeleton-associated, localizing to axons, dendrites, and the cell membrane; it can also be secreted.
  • Tissue expression is largely neuronal, with CNS-specific isoforms and a peripheral nervous system-specific isoform (PNS-tau).
  • Tau is heavily post-translationally modified, including phosphorylation, acetylation, glycation, and ubiquitination, which modulate its function and pathological aggregation.
  • Dysfunction and aggregation of hyperphosphorylated tau are hallmarks of Alzheimer's disease and other tauopathies.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic phosphorylated peptide around T205, so this antibody specifically recognizes tau phosphorylated at this site. For WB, include phosphatase inhibitors during sample preparation to preserve phosphorylation.
  • Tau has a predicted molecular weight of ~79 kDa, but splice isoforms and phosphorylation may result in bands between 48–68 kDa; validate using appropriate positive controls and molecular weight markers.
  • This antibody has been validated for Western blot (WB) and ELISA. For other applications, optimization is recommended.
  • Cross-reactivity with mouse and rat is reported; alignment of the T205 region shows high conservation, but verify sequence similarity if using other species.
