Biological Background: Tau Function and Localization
- Tau (microtubule-associated protein tau) is encoded by the MAPT gene and exists in multiple isoforms due to alternative splicing.
- Promotes microtubule assembly and stability, and plays a critical role in establishing and maintaining neuronal polarity. Related references: PMID:21985311, PMID:32961270
- Functions as a linker between axonal microtubules and neural plasma membrane components via its distinct C- and N-terminal domains. Related references: PMID:21985311, PMID:32961270
- Predominantly localized in the cytoplasm, including the cytosol and cytoskeleton; also found at the cell membrane, in axons, dendrites, and can be secreted.
- Subject to various post-translational modifications including phosphorylation, acetylation, methylation, glycation, and ubiquitination, which regulate its function and aggregation.
- Expressed in neurons, with the PNS-tau isoform specific to the peripheral nervous system and other isoforms in the central nervous system.
- Hyperphosphorylated tau aggregates into paired helical filaments (PHF-tau) forming neurofibrillary tangles, a hallmark of Alzheimer's disease and other tauopathies.
Experimental Guidance and Technical Tips
- This antibody was raised against a synthetic phosphopeptide corresponding to Ser396 phosphorylation site of human tau; verify specificity using non-phosphorylated peptide or phosphatase-treated samples.
- For Western blot, the predicted molecular weight is ~79 kDa, but multiple isoforms and post-translational modifications may produce additional bands; optimize gel percentage and transfer conditions.
- In ELISA, consider using a phospho-specific detection system with appropriate controls to ensure signal specificity.
- Cross-reactivity has been observed with mouse tau; validate in the relevant sample system if using mouse models.
