Biological Background: SYK Function and Localization
- SYK (Spleen tyrosine kinase) is a non-receptor tyrosine kinase that mediates signal transduction downstream of a variety of transmembrane receptors, regulating innate and adaptive immunity, cell adhesion, osteoclast maturation, platelet activation and vascular development. Related references: PMID:12387735 PMID:33782605
- Essential for B-cell receptor (BCR) signaling, SYK is necessary for the maturation of B-cells at the pro-B to pre-B transition. Related references: PMID:12456653
- Upon BCR engagement, SYK phosphorylates key adapters such as BLNK and effectors like PLCG1, linking the activated receptor to calcium mobilization and PKC signaling pathways.
- In the innate immune system, SYK is activated by C-type lectin receptors (e.g., CLEC7A) upon fungal stimulation, triggering ROS production, inflammasome activation, and NF-κB-mediated cytokine transcription.
- SYK is required for IL-15-stimulated neutrophil phagocytosis, highlighting its role in myeloid cell function. Related references: PMID:15123770
- Subcellularly, SYK localizes at the cell membrane, in the cytoplasm, and in the cytosol, dynamically translocating to activated receptors.
- Widely expressed in hematopoietic cells, with prominent expression in B-cell lineages (from pro-B to plasma cells) and neutrophils. Related references: PMID:8163536
- Post-translational modifications include autophosphorylation at tyrosine residues 525 and 526 in the activation loop, as well as ubiquitination, which regulate kinase activity and protein stability.
Experimental Guidance and Technical Tips
- Because this antibody specifically recognizes phosphorylation at Tyr525 and Tyr526, ensure that cell lysates are prepared with phosphatase inhibitors to preserve phosphorylation. For Western blot, validate the signal by treating samples with lambda phosphatase or by comparing stimulated vs. unstimulated conditions.
- When performing ELISA, consider using a phospho-peptide coating antigen and a non-phosphorylated control peptide to confirm phosphorylation specificity.
- Optimal antibody dilutions for WB should be determined empirically; typical starting dilutions for polyclonal antibodies range from 1:500 to 1:2000.
- Cross-reactivity with other species is not expected based on immunogen design, but has not been experimentally verified.
