Biological Background: SYK Function and Localization
- SYK (Spleen tyrosine kinase) is a non-receptor tyrosine kinase that mediates signal transduction downstream of various immunoreceptors, including the B-cell receptor (BCR), through assembly into signaling complexes at the plasma membrane via SH2 domain interactions with phosphorylated ITAM domains. Related references: PMID:12387735, PMID:33782605
- Essential for BCR signaling, SYK phosphorylates downstream effectors such as BLNK, PLCG1, and BTK, and is required for B-cell maturation at the pro-B to pre-B transition. Related references: PMID:12456653, PMID:15388330, PMID:34634301
- Functions in cell adhesion and integrin signaling, regulating neutrophil and macrophage activation, as well as platelet activation and adhesion via collagen and fibrinogen receptors. Related references: PMID:12387735, PMID:15123770
- Widely expressed in hematopoietic cells, including B-cells, neutrophils, and platelets, consistent with its roles in innate and adaptive immunity. Related references: PMID:8163536, PMID:15123770
- Subcellular localization is primarily at the cell membrane and in the cytoplasm, with reports in the cytosol; post-translational modifications include phosphorylation and ubiquitin conjugation.
- Keywords associated with SYK include kinase, SH2 domain, tyrosine-protein kinase, and immunity, highlighting its central signaling role.
Experimental Guidance and Technical Tips
- This antibody recognizes the phosphorylated tyrosine residues Y352 of human SYK and Y319 of human ZAP70. To preserve phosphorylation, use phosphatase inhibitors during sample preparation.
- For Western blot, expected bands may appear around 72 kDa (SYK) and 70 kDa (ZAP70), with possible additional bands at 69 kDa, 55 kDa, and 35 kDa corresponding to isoforms or cleavage products; validate in appropriate cell lines (e.g., Ramos B cells, Jurkat T cells) after stimulation.
- For ELISA, consider using a phosphorylated peptide as a positive control and validate in the relevant sample system.
- Due to high homology between SYK and ZAP70 in the phosphorylated region, verify specificity using knockout or knockdown approaches.
