Biological Background: STAT2 Function and Localization

  • STAT2 is a signal transducer and activator of transcription that mediates signaling by type I interferons (IFN-α/β). Upon IFN binding, receptor-associated JAKs (TYK2 and JAK1) phosphorylate STAT2 at tyrosine 690, enabling its dimerization with STAT1 and association with IRF9 to form the ISGF3 transcription complex. Related references: PMID:23391734, PMID:9020188
  • The ISGF3 complex translocates to the nucleus and binds to the interferon-stimulated response element (ISRE), driving the transcription of interferon-stimulated genes (ISGs) that establish an antiviral cellular state. Related references: PMID:23391734, PMID:9020188
  • STAT2 also exerts negative feedback on type I IFN signaling by recruiting USP18 to the IFNAR2 receptor subunit, attenuating the antiviral response. Related references: PMID:28165510
  • Beyond its canonical role, STAT2 regulates mitochondrial fission by modulating the phosphorylation of DNM1L at Ser-616 (activating) and Ser-637 (inactivating), impacting mitochondrial dynamics. Related references: PMID:23391734, PMID:26122121
  • STAT2 primarily localizes to the cytoplasm but shuttles to the nucleus upon activation.
  • The protein contains an SH2 domain critical for phosphotyrosine-mediated interactions and DNA-binding activity, and is subject to ubiquitin conjugation and proteasomal regulation.

Experimental Guidance and Technical Tips

  • This antibody specifically recognizes STAT2 phosphorylated at Tyr690; include phosphatase inhibitors during sample preparation to preserve the phosphorylation signal.
  • For Western blotting, expect a band at approximately 98 kDa; validate in the relevant human or rat sample system.
  • The rabbit monoclonal origin provides high specificity; use appropriate anti-rabbit secondary antibodies for detection.
  • For ELISA applications, consider pairing this antibody with a capture or detection reagent after thorough cross-reactivity testing.
