Biological Background: SMAD2 Function and Localization
- SMAD2 (SMAD family member 2), also known as JV18-1, MADH2, or hMAD-2, is a receptor-regulated SMAD (R-SMAD) that functions as an intracellular signal transducer and transcriptional modulator downstream of TGF-beta and activin type 1 receptor kinases.
- Upon ligand binding, SMAD2 becomes phosphorylated and forms a complex with SMAD4. This complex translocates to the nucleus, binds to the TRE element in target gene promoters, and activates transcription, thereby regulating diverse cellular processes.
- SMAD2 positively regulates PDPK1 kinase activity by promoting its dissociation from the inhibitory 14-3-3 protein YWHAQ, thereby influencing downstream signaling pathways.

- In immunohistochemistry (IHC-P), optimize antigen retrieval conditions as phosphorylation-specific antibodies can be sensitive to fixation methods.
