Biological Background: SMAD2 Function and Localization
- SMAD2 (Mothers against decapentaplegic homolog 2) is a receptor-regulated SMAD (R-SMAD) and intracellular signal transducer of the TGF-beta and activin pathways.
- Upon ligand binding, type 1 receptor kinases phosphorylate SMAD2 at S465/S467, enabling complex formation with SMAD4 and nuclear translocation to regulate target gene transcription.
- Alternative names include JV18-1, hMAD-2, MADH2, and MADR2; the protein belongs to the SMAD family and is encoded by the SMAD2 gene.
- SMAD2 binds the TRE promoter element and, with SMAD4, activates TGF-beta-responsive genes involved in processes such as odontoblastic differentiation.
- Acetylation, phosphorylation, and ubiquitination modifications dynamically regulate SMAD2 activity, stability, and subcellular redistribution.

- The immunogen is a synthetic phosphorylated peptide spanning S465 and S467 of human SMAD2. To detect endogenous phosphorylation, treat cells with TGF-β1 or activin A and include phosphatase inhibitors in lysis buffers.
- For Western blotting, a band around 52 kDa is expected; validate the signal using phosphatase treatment or non-phospho controls.
- The antibody cross-reacts with human and mouse SMAD2; verify species reactivity in your model system.
- For ELISA, consider coating plates with phospho-peptide and non-phospho-peptide to assess specificity for the phosphorylated epitopes.
