Biological Background: Smad2 Function and Localization
- SMAD2 (also known as JV18-1, MADH2, hMAD-2) is a receptor-regulated SMAD (R-SMAD) and key intracellular signal transducer in the TGF-beta and activin signaling pathways.
- Upon ligand binding, SMAD2 is phosphorylated by type 1 receptor kinases, forms a heteromeric complex with SMAD4, translocates to the nucleus, and binds the TRE element to regulate transcription of target genes.
- SMAD2 positively regulates PDPK1 kinase activity by promoting its dissociation from the inhibitory 14-3-3 protein YWHAQ.
