Biological Background: Retinoblastoma-associated protein Function and Localization
- Tumor suppressor that regulates G1/S transition: hypophosphorylated RB1 binds E2F transcription factors, preventing transcription of E2F-responsive genes; cyclin/CDK-dependent phosphorylation (including at Ser807/Ser811) induces dissociation from E2Fs, activating transcription. - Promotes G0-G1 transition upon phosphorylation and activation by CDK3/cyclin-C. Related references: PMID:15084261
- Directly involved in heterochromatin formation, maintaining overall chromatin structure and stabilizing histone methylation; recruits histone methyltransferases SUV39H1, KMT5B, and KMT5C, leading to epigenetic transcriptional repression, including control of histone H4 'Lys-20' trimethylation.
