Biological Background: RB1 Function and Localization

  • Retinoblastoma-associated protein (RB1) is a key tumor suppressor and cell cycle regulator, controlling the G1/S transition and G0-G1 progression. Its activity is governed by phosphorylation status.
    Related references: PMID:10499802 PMID:15084261
  • Hypophosphorylated RB1 binds and inhibits E2F transcription factors, blocking transcription of genes required for S phase entry. It both physically masks the E2F transactivation domain and recruits chromatin-modifying repressors.
    Related references: PMID:10499802
  • RB1 directly contributes to heterochromatin formation and maintenance by stabilizing histone methylation, notably recruiting SUV39H1 and KMT5B/C to methylate H4K20 and enforcing constitutive heterochromatin.
  • The protein also mediates transcriptional repression through SMARCA4/BRG1 by tethering histone deacetylase (HDAC) complexes to promoters such as c-FOS. In neurons, this repression is relieved by calcium-dependent dephosphorylation of RB1.
  • Viral oncoproteins (SV40 large T antigen, HPV E7, adenovirus E1A) target RB1, dissociating the RB1-E2F complex to drive host cell entry into S phase.
  • Subcellularly, RB1 is primarily nuclear, where it exerts its transcriptional regulatory functions, but it can also localize to the cytoplasm.
  • Expressed in retina and foreskin keratinocytes, with additional expression across many tissue types.
    Related references: PMID:20940255
  • Post-translational modifications include extensive phosphorylation (e.g., at S780 by cyclin-dependent kinases), acetylation, and methylation, which collectively modulate its activity, protein interactions, and stability. Keywords include chromatin regulator, tumor suppressor, and host-virus interaction.

Experimental Guidance and Technical Tips

  • This phospho-specific rabbit monoclonal antibody targets the S780 phosphorylation site on RB1; ensure sample phosphorylation status by treating cells with appropriate stimuli (e.g., serum stimulation, CDK4/6 inhibitors) and always include phosphatase inhibitors during lysis to preserve the epitope.
  • For Western blot, the expected band is ~106 kDa. Validate specificity using a non-phospho peptide blocking control, and compare signals in phosphatase-treated versus untreated lysates.
  • In ELISA, consider using synthetic phospho- and non-phospho peptides as coating antigens to confirm phosphorylation-state selectivity.
  • Cross-reactivity is reported for human and rat; if using mouse samples, verify sequence homology around S780 and perform a pilot experiment.
  • Optimize antibody dilution empirically, as no validated working concentration is provided, and always include positive (e.g., proliferating cancer cell lines) and negative (e.g., RB1-null or serum-starved) controls.
