Biological Background: Retinoblastoma-associated protein (RB1) Function and Localization

  • Tumor suppressor protein that regulates the G1/S transition: hypophosphorylated RB1 binds and inhibits E2F transcription factors, repressing cell cycle progression genes. Cyclin/CDK-mediated phosphorylation releases E2Fs to activate S-phase entry (PubMed:10499802). Related references: PMID:10499802
  • Also promotes the G0-G1 transition through phosphorylation by CDK3/cyclin-C (PubMed:15084261). Related references: PMID:15084261
  • Directly involved in heterochromatin formation by maintaining chromatin structure and stabilizing histone methylation; recruits histone methyltransferases SUV39H1, KMT5B and KMT5C to control H4K20 trimethylation.
  • Subcellular localization: Found predominantly in the nucleus, but can shuttle to the cytoplasm.
