Biological Background: Protein-tyrosine kinase 2-beta Function and Localization

  • Non-receptor protein-tyrosine kinase that regulates reorganization of the actin cytoskeleton, cell polarization, migration, adhesion, spreading and bone remodeling.
  • Plays a role in the humoral immune response, being required for normal levels of marginal B-cells in the spleen and for migration of splenic B-cells; also regulates macrophage polarization and T-cell responses.
  • Promotes osteoclastic bone resorption via cooperation with SRC kinase, and may inhibit osteoprogenitor cell differentiation.
  • Functions downstream of a wide range of receptors, including integrin, immune, G protein-coupled, cytokine, chemokine and growth factor receptors, and mediates responses to cellular stress.
  • Upon activation, forms signaling complexes with SRC and SRC family kinases, leading to phosphorylation of additional tyrosine residues that create docking sites for scaffold proteins effectors and substrates.
  • Activates multiple downstream pathways, including PI3K/AKT1, MAP kinase cascades (ERK, JNK), and Rho family GTPases (RHOA, RAC1), thereby controlling diverse cellular processes.
  • Contributes to the regulation of NMDA receptor ion channel activity and intracellular calcium levels by promoting SRC-mediated phosphorylation of NMDA receptors.
  • Most abundant in the brain, particularly in amygdala and hippocampus; also expressed in spleen, lymphocytes, and at lower levels in kidney. Subcellularly, it localizes to the cytoplasm, focal adhesions, lamellipodia, cell cortex, and nucleus.
