Biological Background: PTEN Function and Localization
- PTEN is a dual-specificity protein phosphatase that dephosphorylates tyrosine-, serine- and threonine-phosphorylated proteins, and also acts as a lipid phosphatase with preference for PtdIns(3,4,5)P3, thereby antagonizing the PI3K-AKT signaling pathway. Related references: PMID:9187108, PMID:9256433, PMID:9616126, PMID:16824732, PMID:26504226, PMID:9593664
- It functions as a tumor suppressor by inhibiting cell cycle progression and promoting apoptosis through its dual phosphatase activity; the nuclear monoubiquitinated form possesses greater apoptotic potential. Related references: PMID:10468583, PMID:18716620
- PTEN regulates cell migration and polarity: unphosphorylated PTEN cooperates with MAGI2 to suppress AKT1 activation, and it dephosphorylates focal adhesion kinase to inhibit cell migration and integrin-mediated spreading. Growth factor stimulation induces PTEN phosphorylation, switching its binding preference to DLC1 and promoting RHOA activation at the posterior of migrating cells. Related references: PMID:11707428, PMID:22279049, PMID:26166433
- In the nervous system, PTEN is involved in adult neurogenesis, controlling newborn neuron integration, dendritic development, and synapse formation. It is recruited to postsynaptic membranes upon NMDA receptor activation and is required for long-term depression (LTD).
- The protein localizes to the cytoplasm, nucleus, PML bodies, dendritic spines, postsynaptic density, and is also secreted.
- Expressed at relatively high levels in all adult tissues, including heart, brain, placenta, lung, liver, muscle, kidney, and pancreas.
- Post-translational modifications include phosphorylation (it is a phosphoprotein), acetylation, and ubiquitination, which regulate its activity and subcellular localization.
- PTEN is implicated in diseases such as autism spectrum disorder and cancer; alternative splicing generates isoforms like PTENbeta.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic phosphorylated peptide around S380 and T383, ensuring specificity for the phosphorylated form. For detection of total PTEN, consider using a pan-PTEN antibody.
- For Western blot (WB), the expected band is ~47 kDa (human PTEN); validate in the relevant cell or tissue lysate, and include phosphatase inhibitors during sample preparation to preserve phosphorylation.
- For immunofluorescence (IF-P) and immunohistochemistry (IHC-P), optimize fixation and permeabilization conditions; include appropriate controls such as peptide competition or phosphatase treatment to confirm phospho-specificity.
- For ELISA, the antibody can be used as a capture or detection reagent; pair with a suitable non-phospho antibody for sandwich ELISA to quantify phospho-PTEN levels.
