Biological Background: PKC zeta Function and Localization
- Protein kinase C zeta (PKC zeta) is an atypical, calcium- and diacylglycerol-independent serine/threonine-protein kinase.
- It functions in the phosphatidylinositol 3-kinase (PI3K) pathway and MAP kinase cascades, playing roles in NF-kappa-B activation, mitogenic signaling, cell proliferation, and cell polarity.
- Required for insulin-dependent activation of AKT3 and contributes to glucose transporter SLC2A4/GLUT4 translocation in adipocytes.
- Participates in TNF-dependent NF-kappa-B activation by phosphorylating IKBKB, and is required for T-helper 2 (Th2) differentiation and allergic airway inflammation.
- Subcellular localization includes cytoplasm, endosomes, cell junctions, and membrane.
- Expressed predominantly in brain, with lower levels in lung, kidney, and testis.
- Involved in late synaptic long-term potentiation in hippocampal CA1 cells and long-term memory maintenance.
- Phosphorylates VAMP2 and LRRK1, regulating intracellular trafficking. Related references: PMID:17313651, PMID:36040231
Experimental Guidance and Technical Tips
- Consider validating phospho-specificity by treating samples with lambda phosphatase prior to WB analysis.
- Use gentle lysis buffers with phosphatase inhibitors to preserve the T560 phosphorylation signal.
- For WB, a band near 68 kDa is expected; use appropriate positive controls such as insulin- or EGF-stimulated cell lysates.
- The antibody’s cross-reactivity with rat suggests utility in rodent model studies; validate in the appropriate species context.
- ELISA protocols may benefit from a phospho-peptide blocking step to confirm specificity.
