Biological Background: PKC (pan) (zeta) Function and Localization

  • PKC alpha (PRKCA) is a calcium-activated, phospholipid- and diacylglycerol-dependent serine/threonine kinase that regulates proliferation, apoptosis, differentiation, migration, and adhesion by phosphorylating key substrates including RAF1, BCL2, and CSPG4.
  • It promotes cell proliferation via RAF1-mediated MAPK/ERK activation and protects glioma cells from apoptosis by inhibiting p53-dependent IGFBP3 expression and phosphorylating BCL2.
  • Subcellular localization spans cytoplasm, membrane, and nucleus; during macrophage differentiation, PKC alpha translocates to the nucleus to support macrophage development, and it relocalizes to lamellipodia to modulate cell motility and desmosomal adhesion.
  • In CD4(+) T cell chemotaxis, PKC alpha phosphorylates DOCK8, triggering dissociation from LRCH1 and activation of CDC42 to drive cell migration (PubMed:28028151). Related references: PMID:28028151
  • Highly expressed in gliomas and breast cancers, PKC alpha acts as a tumor promoter and contributes to malignant phenotypes.
