  • Protein kinase C (PKC) family members are serine/threonine kinases activated by calcium, diacylglycerol (DAG), and phospholipids, regulating diverse cellular processes including proliferation, apoptosis, differentiation, and migration.
  • PKC alpha (PRKCA), a widely expressed and representative isoform, is involved in both positive and negative regulation of the cell cycle and can promote cell growth via RAF1/ERK signaling or induce growth arrest through CDK inhibitors.
  • Exhibits anti-apoptotic functions by phosphorylating BCL2 and suppressing p53/TP53-mediated activation of IGFBP3, and is highly expressed in many cancers where it can act as a tumor promoter.
  • Undergoes stimulus-dependent translocation between cytoplasm, cell membrane, mitochondrial membrane, and nucleus, enabling context-specific signaling.
  • In cardiac function, PKC alpha regulates cardiomyocyte hypertrophy, myocardial contractility, and heart failure progression; it phosphorylates cardiac troponin T (TNNT2) to modulate calcium sensitivity.
