Biological Background: PKA C-alpha (PRKACA) Function and Localization
- PKA C-alpha (PRKACA) is the catalytic subunit of cAMP-dependent protein kinase, with ubiquitous isoform 1 and sperm-specific isoform 2, the latter enriched in pachytene spermatocytes.
- It phosphorylates a vast array of substrates in the cytoplasm and nucleus, including histone H1.4, NFKB1, CDC25B, ABL1, VASP, and GLI transcription factors, thereby controlling processes such as transcriptional regulation, cell cycle progression, and platelet aggregation. Experimental Guidance and Technical Tips
- The antibody targets phosphorylated S339 of PRKACA. To confirm specificity for the phosphorylated form, consider using phosphatase-treated cell lysates or competitive blocking with the non-phosphorylated peptide antigen.
- For Western blotting, a band at approximately 41 kDa is expected in human samples. Optimization of blocking conditions and antibody dilution is recommended to reduce background.
