Biological Background: PAK2 Function and Localization
- Serine/threonine-protein kinase PAK2, also known as PAK65, Gamma-PAK, or p58, is a member of the p21-activated kinase (PAK) family.
- Acts as a downstream effector of the small GTPases CDC42 and RAC1; binding leads to autophosphorylation and kinase activation. Related references: PMID:7744004
- Phosphorylates diverse substrates including histone H4 to promote nucleosome assembly, JUN for EGF-induced proliferation, and MAPK4/6 to regulate F-actin polymerization. Related references: PMID:21724829, PMID:21177766, PMID:21317288
- Caspase-mediated cleavage generates PAK-2p34, which translocates to the nucleus and promotes apoptosis via JNK signaling; also phosphorylates MKNK1 to reduce cellular translation. Related references: PMID:12853446, PMID:16617111, PMID:9171063, PMID:15234964
- Localizes to the cytoplasm, nucleus, perinuclear region, and membrane, and exerts kinase-independent functions in mitotic spindle orientation when associated with ARHGEF7 and GIT1. Related references: PMID:19273597, PMID:19923322
- Ubiquitously expressed with higher levels in skeletal muscle, ovary, thymus, and spleen; post-translationally modified by acetylation, myristoylation, and ubiquitin conjugation.
- Also phosphorylates and inhibits CASP7, preventing apoptosis. Related references: PMID:21555521, PMID:27889207
Experimental Guidance and Technical Tips
- Since the immunogen is a synthetic phosphorylated peptide around serine 20, this antibody is expected to specifically recognize PAK2 phosphorylated at S20; consider using phosphatase inhibitors in sample preparation to preserve phosphorylation.
- For Western blot, validation in human cell lines expressing PAK2 and confirm band at ~58 kDa; cross-reactivity with mouse and rat allows studies in these species, but sequence homology around S20 should be checked.
- In ELISA, use a non-phosphorylated peptide control to verify phospho-specificity.
- Include appropriate controls (e.g., untreated vs. phosphatase-treated lysates) to confirm phospho-dependence.
- Optimal working dilutions must be determined empirically; start with manufacturer's recommendations if available.
