Biological Background: p70 S6 Kinase 1 (RPS6KB1) Function and Localization
- Ribosomal protein S6 kinase beta-1, encoded by RPS6KB1, is also known as p70 S6 kinase alpha (p70S6K1), S6K1, STK14A, and p70 S6KA. Multiple isoforms arise via alternative splicing and alternative initiation.
- Functions as a Ser/Thr kinase downstream of mTORC1, phosphorylating key translation regulators including RPS6, EIF4B, and EEF2K to drive cap-dependent protein synthesis (PubMed:11500364, PubMed:12801526, PubMed:14673156). Related references: PMID:11500364 PMID:12801526 PMID:14673156
- Promotes cell growth, proliferation, and cell cycle progression by integrating signals from growth factors and nutrient availability (PubMed:12801526, PubMed:14673156). Related references: PMID:12801526 PMID:14673156
- In the absence of mitogens, inactive p70S6K1 associates with the EIF3 complex; mTORC1-mediated phosphorylation triggers dissociation and activation (PubMed:16286006). Related references: PMID:16286006
- Contributes to cell survival by phosphorylating and inhibiting the pro-apoptotic factor BAD (By similarity), and mediates TNF-induced insulin resistance via phosphorylation and degradation of IRS1 (PubMed:18952604). Related references: PMID:18952604
- Localizes to the cytoplasm, nucleus, mitochondrial outer membrane, and synaptic compartments, enabling regulation of translation, apoptosis, and cytoskeletal dynamics.
- Subject to extensive post-translational control: key phosphorylation sites include Thr389 (mTORC1), Thr229 (PDK1), and Ser371 within the linker region, along with acetylation events. (Keywords: Phosphoprotein, Acetylation, ATP-binding, Nucleotide-binding).
- Widely expressed across human tissues, underscoring its fundamental role in cellular homeostasis.
Experimental Guidance and Technical Tips
- Because this antibody is raised against a phospho-Ser371 peptide, we recommend including phosphatase inhibitors in lysis buffers to preserve phosphorylation during sample preparation.
- Validate antibody specificity by treating cell lysates with lambda phosphatase or by performing peptide competition assays using both phosphorylated and non-phosphorylated Ser371 peptides.
- In WB, expect a major band at approximately 59 kDa for endogenous p70S6K1; additional bands may represent isoforms or other cross-reactive species; confirm identity with appropriate controls.
- This antibody has been reported to cross-react with mouse p70S6K1; for mouse studies, verify performance using mouse tissue or cell lysates.
- For ELISA applications, consider coating with the immunogen peptide or a recombinant phospho-Ser371 protein fragment and titrate the antibody to determine optimal working concentration.
