Biological Background: NFKB1 Function and Localization
- NFKB1 gene encodes the p105 precursor protein, which is subsequently processed to produce the active p50 subunit (Nuclear factor NF-kappa-B p50) (PubMed:1423592). Related references: PMID:1423592
- The p105 precursor also serves as a cytoplasmic retention protein for other NF-kB family members, sequestering them in the cytoplasm until activation signals are received.
- The p50 subunit forms heterodimers with RELA/p65 to constitute the canonical NF-kB p65-p50 complex, which binds to the kB consensus sequence 5'-GGRNNYYCC-3' and activates transcription of target genes (PubMed:1740106, 7830764). Related references: PMID:1740106 PMID:7830764
- NF-kB is a pleiotropic transcription factor involved in inflammation, immune response, cell proliferation, differentiation, and apoptosis, acting as a central mediator of stress and pathogen-induced signaling.
- The inactive NF-kB complex is retained in the cytoplasm by IkB inhibitors; upon stimulation, IkB is phosphorylated by IKK kinases and degraded, releasing NF-kB to translocate into the nucleus and regulate gene expression.
- NFKB1 is subject to extensive post-translational modifications, including phosphorylation, acetylation, ubiquitination, and hydroxylation, which modulate its stability, DNA-binding activity, and transcriptional competence.
- Subcellular localization: Found in both cytoplasm and nucleus, with shuttling between these compartments tightly controlled by stimulus-dependent signaling pathways.
- Alternatively spliced isoforms and multiple family members (RELA, RELB, REL, NFKB2) generate diverse dimer combinations with distinct functions, from transcriptional activation to repression.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic phospho-peptide corresponding to residues surrounding Ser932 of human NFKB1; this antibody is designed to recognize NFKB1 only when phosphorylated at this site.
- For Western blot, include phosphatase inhibitors during sample preparation to preserve phosphorylation status, and consider comparing stimulated vs. unstimulated cell lysates to confirm phospho-specificity.
- Predicted molecular weight of the p105 precursor is ~105 kDa, while the processed p50 subunit migrates at ~50 kDa; both bands may be detected depending on sample conditions.
- This antibody has been validated for Western blot in human, mouse, and rat samples; other applications have not been tested and should be optimized by the end user.
