Biological Background: NFKB1 Function and Localization
- NFKB1 encodes the p105 precursor protein, which is processed into the active p50 subunit of the NF-kappa-B transcription factor complex. The p105 form also retains NF-kappa-B proteins in the cytoplasm. Related references: PMID:1423592
- The NF-kappa-B complex is a pleiotropic transcription factor present in almost all cell types and regulates genes involved in inflammation, immunity, differentiation, cell growth, and apoptosis.

- The immunogen is a synthetic phosphopeptide around S337; confirm specificity by using phosphatase-treated samples or competing with non-phosphorylated peptide.
- For Western blot, consider using samples from cells stimulated to activate NF-kappa-B signaling (e.g., with TNF-α) to observe phospho-specific band shifts at 85kDa/105kDa.
- ELISA detection may be optimized by pairing with a total NFKB1 antibody or using phospho-specific detection.
- Validate in human cell lines or tissues with known NF-kappa-B activation; cross-reactivity with other species is not guaranteed.
