Biological Background: NF-κB2 p100/p52 Function and Localization

  • NF-κB2 encodes the p100 precursor protein, which is proteolytically processed to generate the active p52 subunit; isoform p49 is also expressed. p100 can retain NF-κB complexes in the cytoplasm.
  • NF-κB is a pleiotropic transcription factor that regulates inflammation, immunity, cell differentiation, growth, tumorigenesis, and apoptosis.
  • p52 forms homo- or heterodimers with other NF-κB/Rel family members (RELA/p65, RELB, NFKB1/p50, REL) and binds to the κB consensus sequence 5'-GGRNNYYCC-3' in target gene enhancers.
  • The heterodimeric RelB-p52 complex functions as a transcriptional activator, while the p52-p52 homodimer acts as a repressor.
  • In the non-canonical NF-κB pathway, MAP3K14-activated CHUK/IKKA homodimer phosphorylates p100 associated with RelB, triggering ubiquitin-dependent processing to p52 and nuclear translocation.
  • NF-κB2 is localized in both the cytoplasm and nucleus, with subcellular compartmentalization and post-translational modifications (phosphorylation, ubiquitination) tightly controlling its activity.
  • p52, in concert with RELB, regulates the circadian clock by repressing the transcriptional activator activity of the CLOCK-BMAL1 heterodimer.
  • Chromosomal rearrangements involving NFKB2 are associated with certain lymphoid malignancies.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic phosphopeptide encompassing Ser866; thus, the antibody is expected to specifically recognize NF-κB2 only when phosphorylated at this residue. Use phosphatase inhibitors during sample preparation to preserve phosphorylation.
  • For Western blotting, the predicted molecular weight of p100 is ~97 kDa; however, multiple bands may be observed due to processing intermediates or degradation. Including appropriate positive and negative controls (e.g., phosphatase-treated lysates) is recommended.
  • For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), optimization of antigen retrieval, permeabilization, and blocking steps may be necessary. The antibody's performance should be validated in the relevant tissue or cell system.
  • In ELISA applications, the phospho-specific antibody can be paired with a capture antibody recognizing total NF-κB2 to quantify phosphorylated protein levels.
  • Cross-reactivity with mouse is reported, but reactivity with other species has not been tested; further validation is advised if using non-human/mouse samples.
