Biological Background: NF-κB p65/RelA Function and Localization
- RELA encodes transcription factor p65 (also known as nuclear factor NF-kappa-B p65 subunit), a central subunit of the NF-κB complex.
- NF-κB is a pleiotropic transcription factor that controls genes in inflammation, immunity, cell growth, differentiation, tumorigenesis, and apoptosis, activated by various stimuli.
- p65 forms homo- or heterodimers with other Rel proteins; the RELA-NFKB1 heterodimer is the most abundant and activates transcription.
- In resting cells, NF-κB is retained in the cytoplasm by IκB inhibitors; upon activation, IκB is degraded, freeing NF-κB to translocate to the nucleus and bind DNA.
- Phosphorylation at serine 536 (S536) in the transactivation domain enhances transcriptional activation and serves as a marker of NF-κB pathway activation.
- p65 localizes to both nucleus and cytoplasm, shuttling in response to signals.
- p65 undergoes multiple post-translational modifications (phosphorylation, acetylation, methylation, ubiquitination, S-nitrosylation) that regulate its function.
