Biological Background: NDRG1 Function and Localization

  • NDRG1 (N-myc downstream-regulated gene 1) is a stress-responsive protein involved in hormone responses, cell growth, and differentiation.
  • Functions as a tumor suppressor in many cell types and is required for p53/TP53-mediated caspase activation and apoptosis.
  • Plays essential roles in cell trafficking, particularly in Schwann cells, and is necessary for peripheral nerve myelin sheath maintenance.
  • Involved in vesicular recycling of CDH1 and TF, and may also function in lipid trafficking; protects cells from spindle disruption damage and regulates microtubule dynamics.
  • Localizes to the cytoplasm, centrosome, nucleus, and cell membrane, consistent with its diverse functions.
  • Post-translationally modified by phosphorylation and acetylation; associated with Charcot-Marie-Tooth disease.
  • Ubiquitously expressed, with highest levels in placental membranes, prostate, kidney, small intestine, and ovary; reduced in adenocarcinomas.

  • For Western blot, use a positive control lysate known to express phosphorylated NDRG1 (T346). Phosphatase treatment can confirm specificity for the phosphorylated form.
