Biological Background: NDRG1 Function and Localization

  • Protein NDRG1 (N-myc downstream-regulated gene 1 protein) is a stress-responsive protein involved in hormone responses, cell growth, and differentiation.
  • Acts as a tumor suppressor and is necessary for p53/TP53-mediated caspase activation and apoptosis.
  • Plays a critical role in Schwann cell trafficking and is required for maintenance and development of peripheral nerve myelin sheath.
  • Involved in vesicular recycling of CDH1 and TF, and may function in lipid trafficking.
  • Protects cells from spindle disruption damage, regulates microtubule dynamics, and maintains euploidy by functioning in the p53-dependent mitotic spindle checkpoint.
  • Subcellular localization includes cytoplasm, cytosol, centrosome, nucleus, and cell membrane.
  • Ubiquitously expressed with highest levels in placental membranes, prostate, kidney, small intestine, ovary, and Schwann cells; reduced expression in adenocarcinomas.
  • Post-translational modifications: phosphorylation at Ser330 and other sites; also acetylated.

  • Raised against a synthetic phosphopeptide corresponding to residues around Ser330 of human NDRG1, this antibody is designed for detection of phosphorylated NDRG1 in Western blot and ELISA applications.
  • For Western blot, consider including both untreated and phosphatase-treated samples to verify phospho-specificity. The predicted molecular weight is 43 kDa; use appropriate molecular weight markers.
  • Validate the antibody in cell lines or tissues known to express NDRG1, such as prostate, kidney, or placental membrane samples, and employ loading controls.
  • For ELISA, consider using phosphorylated and non-phosphorylated peptide competition to assess binding specificity.
