Biological Background: MST1/MST2 Function and Localization

  • STK3, also known as MST2, is a serine/threonine-protein kinase that functions as a stress-activated, pro-apoptotic kinase. Upon caspase cleavage, it translocates to the nucleus and induces chromatin condensation and DNA fragmentation. Related references: PMID:11278283, PMID:8566796, PMID:8816758
  • STK3/MST2 is a core kinase of the Hippo signaling pathway, which regulates organ size and tumor suppression by limiting cell proliferation and promoting apoptosis. Together with STK4/MST1, it phosphorylates and activates LATS1/2 kinases, leading to YAP1 and TAZ inhibition. Related references: PMID:15688006, PMID:16930133, PMID:23972470
  • STK3/MST2 is a member of the GC kinase group of STE20-like kinases and shares structural and functional similarities with STK4/MST1. Both kinases are required to repress hepatocyte proliferation and prevent tumor formation.
  • The kinase localizes to the cytoplasm, nucleus, and centrosome. Its centrosomal localization is important for regulating NEK2-mediated centrosome disjunction. Related references: PMID:21076410, PMID:21723128
  • STK3/MST2 undergoes post-translational modifications including phosphorylation at Thr180 (equivalent to Thr183 in STK4/MST1), acetylation, and ubiquitination. This antibody specifically detects the activated phosphorylated forms.
  • STK3 is expressed at high levels in adult kidney, skeletal muscle, and placenta, with lower expression in heart, lung, and brain.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic phosphorylated peptide around Thr180/Thr183; include phosphatase inhibitors during sample preparation to preserve phosphorylation.
  • For Western blot, validate in relevant cell lines or tissue lysates with known MST1/2 pathway activation. Use loading controls and non-phosphorylated peptide blocking as specificity controls.
  • Cross-reactivity with human, mouse, and rat makes this antibody suitable for comparative studies. Confirm target identity via siRNA knockdown or specific inhibitors.
  • For ELISA, optimize coating and detection conditions using recombinant phosphorylated MST1/2 proteins as standards. Consider use as a capture antibody with appropriate detection pairs.
