Experimental Guidance and Technical Tips
- The immunogen is a synthetic phosphorylated peptide spanning T180/Y182 of human MAPK14/MAPK11. Due to high homology among p38 family members in the activation loop, the antibody may cross-react with phosphorylated forms of MAPK12 and MAPK13. Validate specificity using relevant knockout or phospho-deficient controls.
- Suitable for Western blot (WB) and ELISA. In WB, expect bands around 38–42 kDa corresponding to phosphorylated MAPK14/p38α, MAPK11/p38β, MAPK12/p38γ, and MAPK13/p38δ under stimulated conditions.
- Use fresh samples with phosphatase inhibitor cocktails to preserve phosphorylation. Avoid repeated freeze-thaw cycles of the antibody and consider using a blocking buffer optimized for phospho-specific antibodies.
