Biological Background: MAP2K4 Function and Localization
- Dual specificity mitogen-activated protein kinase kinase 4 (MAP2K4), also known as JNKK1, MEK4, MKK4, SEK1, SKK1, SERK1, MAPKK4, PRKMK4, SAPKK1.
- Essential component of the stress-activated protein kinase/c-Jun N-terminal kinase (SAP/JNK) signaling pathway, directly activating JNK1, JNK2, and JNK3.
- Together with MAP2K7, MAP2K4 is one of only two kinases that directly phosphorylate and activate JNKs; MAP2K4 preferentially phosphorylates the tyrosine residue in the Thr-Pro-Tyr motif, while MAP2K7 targets the threonine.
- Also activates p38 MAPKs (MAPK11, MAPK12, MAPK13, MAPK14), distinguishing it from MAP2K7 which exclusively activates JNKs.
- Involved in mitochondrial death signaling, promoting cytochrome c release and apoptosis.
- Subcellular localization is primarily cytoplasmic, with some nuclear presence.
- Widely expressed with particularly abundant expression in skeletal muscle.
- Post-translational modifications include phosphorylation at S257 and other sites, acetylation, and methylation.

- The immunogen is a synthetic phosphopeptide around S257; the antibody is expected to recognize phosphorylated MAP2K4 at this site. For Western blot, a band at approximately 44 kDa is anticipated.
- Include phosphatase inhibitors during sample preparation to preserve phosphorylation. Validate specificity using non-phosphorylated peptide competition or phosphatase treatment.
- For IHC-P and IF/ICC, optimize antigen retrieval and consider using phospho-specific and total protein controls to confirm localization.
