Biological Background: MAP2K4 Function and Localization

  • Dual specificity mitogen-activated protein kinase kinase 4 (MAP2K4), also known as JNKK1, MEK4, MKK4, SEK1, SKK1, SERK1, MAPKK4, PRKMK4, SAPKK1.
  • Essential component of the stress-activated protein kinase/c-Jun N-terminal kinase (SAP/JNK) signaling pathway, directly activating JNK1, JNK2, and JNK3.
  • Together with MAP2K7, MAP2K4 is one of only two kinases that directly phosphorylate and activate JNKs; MAP2K4 preferentially phosphorylates the tyrosine residue in the Thr-Pro-Tyr motif, while MAP2K7 targets the threonine.
  • Also activates p38 MAPKs (MAPK11, MAPK12, MAPK13, MAPK14), distinguishing it from MAP2K7 which exclusively activates JNKs.
  • Involved in mitochondrial death signaling, promoting cytochrome c release and apoptosis.
  • Subcellular localization is primarily cytoplasmic, with some nuclear presence.
  • Widely expressed with particularly abundant expression in skeletal muscle.
