Biological Background: Dual specificity mitogen-activated protein kinase kinase 3 Function and Localization
- Encoded by the MAP2K3 gene, this protein is also known as MAPK/ERK kinase 3 (MEK3), stress-activated protein kinase kinase 2 (SAPKK2), and MKK3.
- Functions as a dual specificity kinase, catalyzing phosphorylation of both threonine and tyrosine residues on the MAP kinase p38 (MAPK14), thereby activating it.
- Activated by cytokines and environmental stress, playing a critical role in the cellular stress response and inflammation signaling.
- Participates in a signaling cascade initiated by the adrenergic receptor ADRA1B, leading to MAPK14 activation.
- Widely expressed across tissues, with particularly abundant expression in skeletal muscle.
- Subject to alternative splicing and post-translational modifications including acetylation and phosphorylation, which regulate its activity and interactions.
- Related kinase MAP2K6 (MKK6) also activates p38 MAP kinases and shares the S207 phosphorylation site recognized by this antibody.
Experimental Guidance and Technical Tips
- Since the immunogen is a synthetic phosphorylated peptide around S189 of MAP2K6/MAP2K3, the antibody is predicted to specifically recognize the phosphorylated forms of these kinases. Validate specificity using phosphatase treatment or non-phosphorylated peptide blocking in Western blot and ELISA.
- For Western blot, consider optimizing antibody dilution and blocking conditions to reduce non-specific binding. The predicted molecular weights are around 36–39 kDa for MAP2K3 and 31–37 kDa for MAP2K6 depending on isoforms.
- This antibody may be suitable for detecting phosphorylation changes upon stimulation. Confirm by treating cells with relevant stressors (e.g., cytokines, osmotic shock) and monitoring band shifts or signal intensity.
- In ELISA, use appropriate phosphorylated peptide coatings and consider competition assays with non-phosphorylated peptides to ensure phospho-specificity.
