Biological Background: LRRK2 Function and Localization

  • LRRK2 (Leucine-rich repeat serine/threonine-protein kinase 2), also known as Dardarin or PARK8, is a large 286 kDa multidomain protein belonging to the ROCO family. It contains leucine-rich repeats, a Roc GTPase domain, a COR domain, a kinase domain, and WD40 repeats.

  • LRRK2 functions as a serine/threonine kinase and a GTPase, phosphorylating a broad range of substrates involved in neuronal plasticity, innate immunity, autophagy, and vesicle trafficking. Related references: PMID:17114044 PMID:20949042 PMID:21850687

  • It is a key regulator of RAB GTPases, phosphorylating multiple RAB proteins (including RAB3, RAB8, RAB10, RAB12, RAB29, RAB35, and RAB43) to control their membrane association and effector interactions, thereby modulating vesicle trafficking and ciliogenesis. Related references: PMID:23395371 PMID:26824392 PMID:28720718

  • LRRK2 positively regulates autophagy via a calcium-dependent CAMKK/AMPK pathway, and it is recruited to overloaded lysosomes where it phosphorylates RAB8A and RAB10 to promote lysosomal content release. Related references: PMID:22012985 PMID:30209220 PMID:38227290

  • Acts as a positive regulator of innate immunity by mediating phosphorylation of RIPK2 downstream of NOD1 and NOD2, thereby enhancing RIPK2 activation. Related references: PMID:27830463

  • Localizes to multiple cellular compartments including cytoplasmic vesicles, Golgi apparatus membrane, endoplasmic reticulum, endosomes, lysosomes, mitochondrial outer membrane, synaptic vesicles, and cell projections such as axons and dendrites.

  • Highly expressed in the brain, particularly in pyramidal neurons, dopaminoceptive areas, and the substantia nigra pars compacta; also expressed in heart, liver, lung, kidney, and immune cells such as neutrophils. Related references: PMID:29127255

  • Mutations in LRRK2 are the most common genetic cause of familial Parkinson's disease, with pathogenic variants clustering in the GTPase and kinase domains.

Experimental Guidance and Technical Tips

  • Since this is a phospho-specific antibody (anti-phospho-S935), ensure to include phosphatase inhibitors during sample preparation to preserve phosphorylation signals.
  • For WB, the high molecular weight (286 kDa) requires adequate separation conditions; consider using low percentage SDS-PAGE gels and extended transfer times.
  • Validate specificity by comparing signals between phosphorylated and non-phosphorylated forms; peptide blocking with the immunizing phospho-peptide can confirm phospho-specificity.
  • Cross-reactivity with mouse and rat LRRK2 (likely the equivalent phospho-site) has been reported; verify sequence homology of the immunogen epitope in these species.
  • For ELISA, typical direct or sandwich formats can be used; titrate antibody concentration in the relevant sample system.
