Biological Background: LRP6 Function and Localization

  • LRP6 (Low-density lipoprotein receptor-related protein 6) is a single-pass transmembrane coreceptor that, together with Frizzled (Fzd), forms the Wnt-Fzd-LRP5-LRP6 complex, which triggers beta-catenin signaling by inducing aggregation into ribosome-sized signalosomes. Related references: PMID:11357136, PMID:11448771, PMID:15778503
  • Upon Wnt activation, the Fzd/LRP6 coreceptor recruits DVL1 polymers and the AXIN1/GSK3B complex to the plasma membrane, forming signalosomes that inhibit AXIN1/GSK3-mediated phosphorylation and destruction of beta-catenin. Related references: PMID:16513652
  • LRP6 localizes to the cell membrane, endoplasmic reticulum, and membrane rafts, consistent with its roles in signal reception and trafficking.
  • Widely coexpressed with LRP5 during embryogenesis and in adult tissues, reflecting its broad involvement in developmental and homeostatic Wnt signaling.
  • Post-translational modifications include phosphorylation at multiple serine/threonine residues (including Ser1490), palmitoylation, and ubiquitination, which regulate receptor activity and stability.
  • Characterized by structural motifs such as EGF-like domains and LDL-receptor class A repeats, shared with other LDL receptor family members.
  • LRP6 is recognized as a disease variant protein; mutations in LRP6 are associated with osteopetrosis and other skeletal disorders.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic phosphorylated peptide around Ser1490 of human LRP6; to confirm phospho-specificity, consider treating cell lysates with lambda phosphatase and comparing signals, or perform peptide competition assays with non-phosphorylated and phosphorylated peptides.
  • For Western blotting, LRP6 migrates at approximately 180 kDa; use appropriate gel percentages, phospho-safe lysis buffers, and protease/phosphatase inhibitors to preserve phosphorylation status.
  • In ELISA, optimize coating conditions and antibody dilutions; pairing with a capture antibody may improve detection sensitivity for endogenous phospho-LRP6 in relevant sample systems.
  • Due to homology with LRP5 and other LDL receptor family members, cross-reactivity testing in the intended sample matrix is recommended to confirm specificity.
