Biological Background: JNK1/JNK2/JNK3 Function and Localization
- Mitogen-activated protein kinase 8 (MAPK8/JNK1) is a serine/threonine-protein kinase belonging to the c-Jun N-terminal kinase (JNK) family, which also includes MAPK9/JNK2 and MAPK10/JNK3. These kinases are encoded by MAPK8, MAPK9, and MAPK10 respectively and share conserved activation loop phosphorylation sites (Thr183 in JNK1/JNK2 and Thr221 in JNK3).
- JNK1 is a central mediator of the stress-activated protein kinase (SAP/JNK) signaling pathway, which is stimulated by pro-inflammatory cytokines and physical stress. Activated JNK1 phosphorylates transcription factors including JUN, JDP2, and ATF2, thereby regulating AP-1 transcriptional activity (PubMed:18307971).

PMID:18307971 - JNK1 promotes apoptosis by phosphorylating p53/TP53 and YAP1 (PubMed:21364637). It also supports erythroid cell survival by phosphorylating BAD upon EPO stimulation (PubMed:21095239) and mediates starvation-induced autophagy through BCL2 phosphorylation (PubMed:18570871).

PMID:21364637, PMID:21095239, PMID:18570871 - JNK1 phosphorylates a broad range of substrates including CDT1 (PubMed:21856198), HSF4 (PubMed:16581800), SIRT1 (PubMed:17296730), ELK1, ITCH (PubMed:20027304), CLOCK-BMAL1 (PubMed:22441692), NLRP3 (PubMed:28943315), and SIRT6 (PubMed:27568560), implicating it in DNA replication, circadian rhythm, inflammasome activation, and cellular stress responses.

- JNK1 localizes to the cytoplasm, nucleus, and synapses, allowing it to regulate both nuclear transcription and cytoplasmic events such as synaptic vesicle capture via SYT4 phosphorylation.
Experimental Guidance and Technical Tips
- The antibody was raised against a synthetic phospho-peptide around Thr183 of human JNK1. This activation‑loop residue (Thr183 in JNK1/JNK2, Thr221 in JNK3) is conserved across the three JNK family members, explaining the multi‑specificity seen in the specifications.
- For Western blotting, use phosphatase inhibitor‑supplemented lysis buffers to preserve the phosphorylation signal. The predicted molecular weights range from 35‑52 kDa, but actual band positions may vary due to isoform expression and post‑translational modifications; validate in the relevant sample system.
- In immunohistochemistry and immunofluorescence, phospho‑specific detection may require antigen retrieval and careful optimization of fixation/permeabilization conditions. Always include a non‑phospho control or phosphatase treatment to verify signal specificity.
- For ELISA applications, consider pairing this antibody with a total‑JNK capture antibody to quantify phosphorylation levels relative to total protein expression.
