Biological Background: IκBα Function and Localization
- NF-κB inhibitor alpha (IκBα) is a key regulator of NF-κB transcription factors, encoded by the NFKBIA gene. It is also known as I-kappa-B-alpha, IkB-alpha, and MAD3.
- IκBα functions by binding to NF-κB dimers (such as RELA/p65 and NFKB1/p50) in the cytoplasm, masking their nuclear localization signals and preventing nuclear translocation. Related references: PMID:1493333, PMID:36651806, PMID:7479976
- Upon pro-inflammatory or immune stimulation, IκBα undergoes phosphorylation at serine residues 32 and 36, leading to ubiquitination and proteasomal degradation, releasing NF-κB to translocate to the nucleus and activate transcription. Related references: PMID:7479976, PMID:7628694, PMID:7796813
- IκBα predominantly localizes to the cytoplasm but can shuttle into the nucleus, where it may also inhibit NF-κB DNA binding or mediate termination of NF-κB response.
- Phosphorylation at Ser32 is a critical event in the canonical NF-κB signaling pathway, making it a widely studied biomarker for pathway activation.
- Keywords from UniProt indicate that IκBα is a phosphoprotein, undergoes ubiquitination, and is involved in host-virus interactions, highlighting its central role in immune signaling and disease.
- The protein has a molecular weight of approximately 35.6 kDa (calculated) and contains ankyrin repeat domains that mediate interactions with NF-κB.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic phosphopeptide around Ser32 of human IκBα; therefore, the antibody specifically recognizes the phosphorylated form. Always include phosphatase inhibitors during sample preparation for Western blot to preserve the phosphorylation signal.
- For ELISA applications, the antibody can be used as a capture or detection reagent for quantifying phospho-IκBα in cell or tissue lysates; consider pairing with a total IκBα antibody for normalization.
- Confirm antibody specificity in your system by treating samples with lambda phosphatase or by using a non-phosphorylated peptide as a competitor.
- Cross-reactivity is reported for human, mouse, and rat. Validate reactivities in the relevant species before large-scale experiments, as epitope conservation may vary.
