Biological Background: Hormone-Sensitive Lipase Function and Localization
- Hormone-sensitive lipase (HSL) is a broad-specificity lipase that catalyzes the hydrolysis of triacylglycerols (TAGs), diacylglycerols (DAGs), monoacylglycerols (MAGs), cholesteryl esters, and retinyl esters. It shows preferential hydrolysis of DAGs over TAGs and MAGs, and preferentially cleaves fatty acid esters at the sn-3 position of DAGs (PubMed:15716583, PubMed:15955102, PubMed:19800417). Related references: PMID:15716583 PMID:15955102 PMID:19800417
- In adipose tissue and heart, HSL primarily hydrolyzes stored triglycerides to release free fatty acids, while in steroidogenic tissues, it converts cholesteryl esters to free cholesterol for steroid hormone production.
- The protein, encoded by the LIPE gene, is also known as monoacylglycerol lipase LIPE, retinyl ester hydrolase (REH), and is implicated in metabolic disorders such as diabetes and obesity.
- HSL localizes to multiple cellular compartments including the cell membrane, caveolae, cytosol, and lipid droplets, consistent with its role in lipid mobilization.
- Phosphorylation at serine 853 (S853) is a key regulatory modification that modulates HSL enzymatic activity and subcellular localization; the protein is classified as a phosphoprotein involved in lipid degradation and cholesterol/steroid metabolism.
- Tissue expression is highest in testis, with additional presence in adipose, heart, and steroidogenic tissues, reflecting its tissue-specific metabolic functions.
Experimental Guidance and Technical Tips
- This antibody is raised against a synthetic phosphopeptide corresponding to phosphorylated serine 853 of human HSL; it specifically recognizes the phosphorylated form of HSL and should not detect the non-phosphorylated protein.
- For Western blotting, use lysis buffers containing phosphatase inhibitors to preserve the phosphorylation signal. HSL runs at approximately 117 kDa; optimize the antibody dilution based on your sample type and detection system.
- Cross-reactivity with mouse is confirmed; thus, the antibody can be used for mouse tissue or cell lysates. Validate specificity in your relevant biological context, such as lipolytic or steroidogenic tissues.
- For ELISA, consider using a phospho-specific coating or detection strategy; verify linearity and sensitivity with known phosphorylated HSL standards or peptide competition assays.
