Biological Background: Tyrosine-protein kinase Fyn Function and Localization
- FYN encodes a non-receptor tyrosine kinase of the Src family, also known as Proto-oncogene Syn, c-Fyn, Src-like kinase (SLK), and p59-Fyn, containing SH2 and SH3 domains.
- FYN is a non-receptor tyrosine-protein kinase that regulates cell growth, survival, adhesion, integrin-mediated signaling, cytoskeletal remodeling, cell motility, immune response, and axon guidance. Related references: PMID:11536198, PMID:15489916, PMID:15557120
- It modulates cell adhesion and motility by phosphorylating CTNNB1 (beta-catenin) and CTNND1 (delta-catenin). Related references: PMID:17194753
- Cytoskeletal remodeling is mediated through phosphorylation of actin regulator WAS and microtubule-associated proteins MAP2 and MAPT. Related references: PMID:14707117, PMID:15536091
- In T-cells, FYN participates in TCR signaling, phosphorylating PTK2B/PYK2, VAV1, and mediating negative feedback via PAG1 and PDCD1. Related references: PMID:22080863, PMID:20028775, PMID:18056706, PMID:32184441, PMID:11005864
- Neuronal functions include phosphorylation of DPYSL2, ARHGAP32, and SNCA, contributing to axon guidance and synaptic regulation. Related references: PMID:11162638, PMID:12788081, PMID:19652227
- In the kidney, it maintains glomerular filtration by phosphorylating slit diaphragm proteins NPHS1, KIRREL1, and TRPC6. Related references: PMID:14761972, PMID:18258597, PMID:19179337
- Subcellularly, FYN localizes to the cytoplasm, nucleus, cell membrane, and perikaryon; isoform 1 is highly expressed in brain, while isoform 2 predominates in T-lymphocytes.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic phosphopeptide corresponding to residues around serine 21 (S21) of human Fyn (NP_002028.1); consider verifying phospho-specificity using non-phosphorylated peptide controls.
- For Western blotting, include phosphatase inhibitors during sample preparation and validate band authenticity by lambda phosphatase treatment.
- In ELISA, test parallel wells coated with phosphorylated and non-phosphorylated peptides to confirm selective binding.
- Cross-reactivity with species other than human has not been evaluated; perform species-specific validation if required.
