Biological Background: FOXO3A Function and Localization
- FOXO3A (forkhead box O3) is a transcription factor of the forkhead family, encoded by the FOXO3 gene (aliases FKHRL1, FOXO2, AF6q21). It binds the consensus DNA motif 5'-[AG]TAAA[TC]A-3'.
- It triggers apoptosis in the absence of survival factors, including neuronal cell death upon oxidative stress.
Related references: PMID:10102273, PMID:16751106 - FOXO3A participates in post-transcriptional regulation of MYC: phosphorylation by MAPKAPK5 promotes induction of miR-34b/c, which bind MYC 3'UTR and inhibit translation.
Related references: PMID:21329882 - In response to metabolic stress, FOXO3A translocates into mitochondria and promotes mtDNA transcription.
Related references: PMID:23283301 - FOXO3A is a key regulator of regulatory T-cell (Treg) differentiation by activating FOXP3 expression.
Related references: PMID:30513302 - The protein dynamically shuttles between the cytoplasm, nucleus, and mitochondrial compartments (matrix/outer membrane) depending on phosphorylation and cellular cues.
- FOXO3A activity is governed by multiple post-translational modifications, including phosphorylation at Ser/Thr residues (e.g., Ser294), acetylation, methylation, and ubiquitination.
- It is widely expressed in human tissues (ubiquitous).
Experimental Guidance and Technical Tips
- The immunogen is a synthetic phosphopeptide encompassing Ser294 of human FOXO3A. This antibody is suitable for detecting phosphorylated FOXO3A (Ser294) in WB and ELISA applications. Species cross-reactivity is predicted with mouse and rat due to sequence conservation, but endogenous expression levels may vary; validate in the relevant sample system.
- For WB, consider that FOXO3A protein weight is ~71 kDa; however, phosphorylation can induce slight mobility shifts. Use phosphatase inhibitors during sample preparation to preserve phospho-signal.
- As FOXO3A phosphorylation at Ser294 is a dynamic event influenced by cellular conditions (e.g., oxidative stress, growth factor signaling), include appropriate positive and negative controls (e.g., stimulated vs. resting cells) in your experiments.
