  • This antibody was raised against a synthetic phosphopeptide corresponding to the Thr‑24 region of human FOXO1. Based on sequence homology, it is predicted to also recognize the phosphorylated Thr‑32 of FOXO3A and Thr‑28 of FOXO4.
  • For Western blot analysis, include phosphatase inhibitors in the lysis buffer to preserve the phosphorylation signal. A band near 70 kDa is expected for FOXO1, with similar sizes for FOXO3A and FOXO4.
  • When performing immunohistochemistry (IHC‑P), consider validating the staining in tissues known to express FOXO1 (e.g., skeletal muscle, liver) and using a dephosphorylated control to confirm phospho‑specificity.
