Biological Background: ERK1/ERK2 Function and Localization
- MAPK1 (ERK2, p42-MAPK) and MAPK3 (ERK1, p44-MAPK) are serine/threonine kinases that function as the core of the MAPK/ERK signaling cascade, regulating cell growth, adhesion, survival, and differentiation through phosphorylation of over 160 substrates. Related references: PMID:35831023

- The immunogen is a synthetic phosphopeptide derived from the activation loop of human ERK2; expect cross-reactivity with the homologous phosphorylated region of ERK1 due to high sequence conservation.
- In WB, typical bands appear at ~42 kDa (phospho-ERK2) and ~44 kDa (phospho-ERK1) upon stimulation; confirm phospho-specificity by comparing with untreated or phosphatase-treated lysates.
- For IF/ICC, consider co-staining with a total ERK1/ERK2 antibody to validate activation status and subcellular translocation.
- Additional bands near 36–43 kDa may represent splice variants, post-translational modifications, or degradation products; validate in the relevant sample system.
- ELISA applications may be suitable for quantitative detection of phosphorylated ERK in cell or tissue lysates; optimize coating and detection conditions accordingly.
