  • In WB, typical bands appear at ~42 kDa (phospho-ERK2) and ~44 kDa (phospho-ERK1) upon stimulation; confirm phospho-specificity by comparing with untreated or phosphatase-treated lysates.
  • For IF/ICC, consider co-staining with a total ERK1/ERK2 antibody to validate activation status and subcellular translocation.
  • Additional bands near 36–43 kDa may represent splice variants, post-translational modifications, or degradation products; validate in the relevant sample system.
  • ELISA applications may be suitable for quantitative detection of phosphorylated ERK in cell or tissue lysates; optimize coating and detection conditions accordingly.
