Biological Background: ERK1/ERK2 Function and Localization

  • MAPK1 (ERK2) and MAPK3 (ERK1) are serine/threonine kinases that serve as essential components of the MAPK/ERK signal transduction cascade, regulating diverse cellular processes including growth, differentiation, and apoptosis.
  • Upon activation, ERK1/2 phosphorylate over 160 substrates, including transcription factors (e.g., ATF2, ELK1, FOS), cytoskeletal elements, apoptosis regulators, and other kinases, thereby controlling gene expression and cellular dynamics (PubMed:35831023). Related references: PMID:35831023
  • Under hypoxic conditions, ERK2 phosphorylates phosphoglycerate kinase PGK1 to promote its mitochondrial targeting and suppress acetyl-CoA formation (PubMed:26942675). Related references: PMID:26942675

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic phosphorylated peptide corresponding to the T202 region of human ERK1. Use phosphatase inhibitors during sample preparation to preserve the phosphorylation signal.
  • For Western blotting, expected bands are observed at approximately 42 kDa (ERK2) and 44 kDa (ERK1). Optimize antibody dilution and incubation conditions for your specific sample.
  • For immunohistochemistry (IHC-P), perform antigen retrieval and validate with positive control tissues expressing high levels of phospho-ERK1/2.
  • This antibody is not suitable for detecting total ERK1/2 levels; use a pan-ERK antibody for total protein normalization.
