Biological Background: EGFR Function and Localization

  • EGFR (Epidermal Growth Factor Receptor), also known as ErbB-1 or HER1, is a transmembrane receptor tyrosine kinase belonging to the ERBB family. An alternatively spliced isoform (isoform 2) may function as an antagonist of EGF action.

  • For western blot, expect a major band around 134 kDa. To confirm phospho-specificity, consider treating cell lysates with lambda phosphatase or performing peptide competition assays using the non-phosphorylated counterpart of the immunogen peptide.
  • In immunofluorescence/ICC, a predominantly membrane-associated staining pattern is anticipated; co-staining with a plasma membrane marker can help confirm localization.
  • For ELISA applications, the antibody may be used for capturing or detecting phosphorylated EGFR; optimize coating and detection conditions empirically.
