Biological Background: DNMT1 Function and Localization
- DNA methyltransferase that methylates CpG residues, with a preference for hemimethylated DNA. Related references: PMID:17200670, PMID:18754681, PMID:21745816, PMID:26070743
- Associates with DNA replication sites during S phase and with chromatin during G2 and M phases to maintain methylation patterns essential for epigenetic inheritance. Related references: PMID:17200670, PMID:21745816
- Responsible for preserving DNA methylation patterns established during development. Related references: PMID:21745816
- DNA methylation is coordinated with histone modifications; also mediates transcriptional repression by direct binding to HDAC2. Related references: PMID:16357870, PMID:10888872
- In association with DNMT3B and CTCFL/BORIS, activates BAG1 gene expression by modulating dimethylation of histone H3 at H3K4 and H3K9. Related references: PMID:18413740
- Forms a corepressor complex required for activated KRAS-mediated promoter hypermethylation and transcriptional silencing of tumor suppressor genes in colorectal cancer cells; also maintains a transcriptionally repressive state in embryonic stem cells. Related references: PMID:24623306
- Localizes to the nucleus and chromosome.
- Undergoes post-translational modifications including phosphorylation at Ser714 and other residues, acetylation, and methylation.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic phospho-peptide spanning Ser714 of human DNMT1; confirm phospho-specificity by phosphatase treatment or peptide competition assays.
- For Western blotting, DNMT1 migrates at ~183 kDa; optimize transfer conditions for high molecular weight detection.
- In immunofluorescence/ICC, expect nuclear staining; co-immunostain with a total DNMT1 antibody to differentiate phosphorylation-specific signals.
- For ELISA, consider using phospho- and non-phospho-peptides as coating antigens to verify binding specificity.
