Biological Background: Cytokeratin 8 (KRT8) Function and Localization
- Keratin 8 (KRT8), also known as cytokeratin-8 (CK-8) and type-II keratin Kb8, is a type II intermediate filament protein encoded by the KRT8 gene in humans.
- Together with its obligate type I partner KRT18, KRT8 forms the keratin intermediate filament network that provides mechanical stability and resilience to simple epithelial cells. Related references: PMID:26823019
- KRT8 is also involved in protecting cells from non-mechanical stress, such as apoptosis.
- It is required for ARHGEF40-mediated actin stress fiber formation and tensional force-induced stress fiber reinforcement. Related references: PMID:26823019
- In striated muscle, KRT8, together with KRT19, may contribute to linking the contractile apparatus to dystrophin at the costameres. Related references: PMID:16000376

- The immunogen is a synthetic phosphopeptide around Ser74; to confirm phospho-specificity, compare signals with and without phosphatase treatment or use non-phosphorylated peptide competition.
- In Western blot, the expected band is at ~54-56 kDa. Validate detection in your sample type and optimize antibody dilution empirically.
- For ELISA, consider using the phosphopeptide as coating antigen and include a non-phosphorylated peptide control.
- Cross-reactivity is reported for human only; if using other species, perform validation due to potential sequence variation.
- Include appropriate loading controls and molecular weight markers for accurate interpretation.
